      目的研究运动及高脂饮食对OLETF大鼠血清肿瘤坏死因子-α(TNF-α)浓度的影响,探讨TNF-α在运动改善机体胰岛素敏感性中所起的作用。 方法对40只自发发病的2型糖尿病模型——OLETF大鼠和15只LETO大鼠进行9周的运动和高脂饮食干预, 9周后用放射免疫法测定每只大鼠血清中TNF-α的浓度。 结果基础饲料饲养的OLETF大鼠非运动组和运动组TNF-α浓度分别为(0.406±0.083)ng/ml和(0.574±0.116)ng/ml;高脂饲料饲养的OLETF大鼠非运动组和运动组TNF-α浓度分别为(0.407±0.097)ng/ml和(0.513±0.098)ng/ml;而基础饲料饲养的LETO大鼠非运动组和运动组TNF-α浓度分别为(0.387±0.087)ng/ml和(0.382±0.077)ng/ml;OLETF大鼠TNF-α浓度运动组明显高于其相对应的非运动组(P<0.01),运动组和非运动组LETO大鼠TNF-α浓度没有明显的差别。 结论运动能使OLETF大鼠TNF-α浓度增加,而对LETO大鼠TNF-α浓度没有明显影响;高脂饮食对OLETF大鼠TNF-α浓度没有明显影响。

      Objective To investigate the effect of exercise and high-fat diet on OLETF rats serum concentration of TNF-α and the mechanism of exercise enhancing insulin sensitivity. MethodsOLETF rat is a spontaneously diabetic animal model and LETO rat a non-diabetic control model. After 9 weeks of exercise and high-fat diet on 40 OLETF rats and 15 LETO rats, the serum concentration of TNF-α was measured by means of radioactive-immune kit. ResultsThe serum level of TNF-α of non-exercise and exercise OLETF rats fed by chow were(0.406±0.083)ng/ml and(0.574±0.116)ng/ml, respectively; for the OLETF rats fed by high-fat diet, the serum level of TNF-α of those intervened with non-exercise and exercise were(0.407±0.097)ng/ml and (0.513±0.098)ng/ml , respectively. For the LETO rats, the serum concentration of TNF-α of those with non-exercise and exercise were(0.387±0.087)ng/ml and(0.382±0.077)ng/ml, respectively. The serum level of TNF-α in exercise OLETF rats were significantly higher than that in non-exercise OLETF rats(P<0.01), however, the difference of the serum level of TNF-α between the non-exercise and exercise LETO rats was not statistically significant. ConclusionExercise can increase the level of serum TNF-α in OLETF rats; and high-fat diet had not changed significantly the serum concentration of TNF-α in OLETF rats.
