      目的研究不同剂量短波紫外线(ultraviolet C, UVC)照射对伤口肉芽组织中碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)表达的影响。 方法将每只实验大鼠背部制作3个皮肤全层伤口,分别采用不同剂量(15 mJ/cm2或60 mJ/cm2)UVC照射大鼠背部伤口,持续3 d后,分别于第7天、第14天及第21天应用免疫组化法及原位杂交法观察伤口肉芽组织中bFGF的表达情况。 结果致伤后7 d,60 mJ/cm2照射组及15 mJ/cm2 照射组bFGF表达均高于对照组(P均<0.01),且以60 mJ/cm2照射组增高的更为显著;致伤后14 d,60 mJ/cm2照射组bFGF表达显著降低,低于15 mJ/cm2照射组及对照组(P均<0.05)。 结论在伤口愈合早期,采用不同剂量(15 mJ/cm2或60 mJ/cm2)UVC照射能明显促进伤口肉芽组织内bFGF的表达,其中剂量为60 mJ/cm2的UVC照射其作用短暂而迅速,而剂量为15 mJ/cm2的UVC照射其作用缓慢而持久。
英文摘要:
      Objective To investigate the influence of ultraviolet C (UVC) irradiation of different doses on expression of basic fibroblast growth factor (bFGF) in granulation tissues of wound in rats. MethodsThirty Wistar rats were recruited and three wounds were made in each rat. The UVC irradiation of different doses (60mJ/cm2 and 15mJ/cm2) were performed daily in two of the three wounds in each rat, respectively, and the UVC irradiation lasted for three days. The expression of bFGF both at the mRNA level and the protein level were observed by the methods of in situ hybridization and immunohistochemistry at 7 days, 14 days, 21 days after wound was made, respectively. ResultsAt the 7 days after wound was made, the expression of bFGF in irradiation groups were higher than that in the control group (P<0.05) at both the mRNA and the protein level, especially the increment in the 60mJ/cm2 group was more significant. While at 14 day after the wound establishment, the expression of bFGF in the 60mJ/cm2 group decreased rapidly and the concentration of bFGF were lower than that in the other two groups (P<0.05). At the same time, the expression of bFGF in the 15mJ/cm2 group was higher than that in the 60mJ/cm2 group. ConclusionIn the initial stage of wound healing, the treatment of UVC irradiation promoted the expression of bFGF and the effect of 60mJ/cm2 irradiation was fast and transit, the effect of the 15mJ/cm2 irradiation was durative and slow. The increment of bFGF promoted by UVC irradiation might be the one of the mechanisms of UVC irradiation healing wound.
