      目的从基因水平探讨TNF-α和TNF-α R1在增生性瘢痕组织中的作用,为增生性瘢痕的基因治疗探索一条有效途径。 方法以正常瘢痕为对照,利用RT-PCR 技术分别检测1年以上增生性瘢痕组织中成纤维细胞TNF-α及TNF-α R1 mRNA的表达情况。 结果在增生性瘢痕组织中,TNF-αmRNA、TNF-αR1mRNA表达的相对值分别为(1.28±0.55)和(1.02±0.13),均明显低于正常瘢痕组织(P<0.05)。 结论提示TNF-α在正常伤口的愈合过程中起着重要的作用,增生性瘢痕的形成可能与组织中TNF-α和TNF-α R1表达低下有关。基因治疗可提高早期增生性瘢痕中TNF-α的含量或靶细胞膜上TNF-α R1的数量与活性,可能成为预防增生性瘢痕发生的一条有效途径。
英文摘要:
      Objective To explore the role of tumor necrosis factor alpha (TNF-α) and its receptor (TNF-α R1) in hypertrophic scar at the gene level. MethodsThe expression of TNF-α mRNA and TNF-α R1 in hypertrophic scars was tested by semiquantitative reverse polymerase chain reation (RT-PCR) method and was compared with that in the normal scars. ResultsThe expression ratios of TNF-α mRNA and TNF-α R1 in hypertrophic scars were (1.28±0.55) and (1.02±0.13), respectively,which were significantly lower than those in the normal ones. ConclusionThe results suggested that TNF-α might play an important role in the process of wound healing and that the formation of hypertrophic scar might be due to the lack of TNF-α or the down-regulation of TNF-α R1. Therefore, it was reasonable to increase the level of TNF-α or up-regulate the amount of TNF-α R1 by using gene therapy in the early stage of hyphertrophic scar development.
