      目的观察姜黄素联合电针治疗对大鼠缺血脑组织中脑源性神经营养因子(BDNF)及神经生长因子(NGF)表达的影响。 方法选取健康雄性Wistar大鼠,采用线栓法将其制成大脑中动脉栓塞大鼠模型,采用随机数字表法将制模成功大鼠分成4组,分别是对照组、电针组、姜黄素组及电针+姜黄素组(简称观察组),每组15只。姜黄素组及电针组大鼠于制模后分别给予姜黄素腹腔注射或电针治疗,观察组大鼠于制模后则联合给予姜黄素腹腔注射及电针治疗,对照组大鼠制模后未给予特殊处理。于制模后14d时对各组大鼠进行神经功能评分,然后处死各组大鼠分别取脑梗死区脑组织制成标本,采用免疫组化染色技术检测各组大鼠脑梗死区BDNF及NGF阳性细胞表达情况。 结果制模后14d时发现观察组大鼠神经功能缺损评分[(1.37±0.59)分]明显低于电针组、姜黄素组及对照组评分[分别为(1.59±0.38)分、(1.68±1.06)分和(1.98±0.26)分],组间差异均具有统计学意义(P<0.05);另外观察组大鼠脑梗死区BDNF及NGF阳性细胞表达均较其他三组明显增强,组间差异均具有统计学意义(P<0.05)。 结论姜黄素联合电针治疗可促进大脑中动脉栓塞大鼠脑缺血组织BDNF及NGF表达,并可能通过该机制发挥神经保护作用。

      Objective To investigate the expression of brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) after cerebral infarction and treatment with curcumin combined with electroacupuncture. MethodsMiddle cerebral artery occlusion was successfully introduced to simulate cerebral infarction in 60 healthy, male Wistar rats. They were then randomly divided into a control group, an electroacupuncture group, a curcumin group and a curcumin combined with electroacupuncture group (the observation group), each of 15. After the modeling, the four groups were given the planned interventions for 2 weeks, with the control group receiving no intervention. All of the rats were sacrificed after neurobehavioral scoring and immunological staining of brain tissue from the infarction area was used to detect the expression of BDNF and NGF. ResultsThe average Bederson score in the observation group was significantly lower than those of the other groups. Cells expressing BDNF and NGF were significantly more numerous in the observation group than in the other three groups. ConclusionCurcumin combined with electroacupuncture can significantly improve the expression of BDNF and NGF in the infarcted area, and this may play a neuroprotective role.
