      目的观察新生大鼠缺氧缺血性脑损伤(HIBI)后,外源性脑源性神经营养因子(BDNF)对脑内源性阿片肽(EOP)水平的影响。 方法7 d龄新生大鼠随机分为HIBI+BDNF组(A组)、单纯HIBI组(B组)和假手术组(C组)。结扎幼鼠左侧颈总动脉并在低氧(8%O2+92%N2)环境下形成HIBI,造模后立刻向侧脑室注射0.5 μg BDNF,给药后即刻、60 min及120 min观察左、右侧皮层和海马强啡肽A1-13免疫活性物质(ir-DynA1-13)、β-内啡肽免疫活性物质(ir-β-EP)和亮啡肽免疫活性物质(ir-LEK)含量的变化。 结果①各组新生大鼠HIBI后大多数时间点相比,B组皮层、海马ir-DynA1-13和ir-β-EP含量较C组显著升高(P<0.05或P<0.01);伤后即刻,B组左侧皮层、海马ir-LEK含量显著高于C组(P<0.05)。②给予BDNF后,A、B组ir-DynA1-13含量在大多数时间点与C组比较,差异有显著性意义(P<0.05或P<0.01);皮层在给药后60 min,A组ir-β-EP含量显著低于B组(P<0.05),其余各时间点与B组比较,差异均无显著性意义;A、C组比较,海马ir-LEK含量差异均无显著性意义。 结论脑内EOP中,以ir-DynA1-13、ir-β-EP与HIBI关系更为密切。通过影响强啡肽与内啡肽系统对抗HIBI是BDNF的作用途径之一。

      Objective To explore the effects of brain-derived neurotrophic factors (BDNF) on concentrations of cerebral endogenous opioid peptides(EOP)in neonatal rats subject to with hypoxic ischemic brain injury (HIBI). MethodsSeven-day-old Sprague Dawley rats were randomly divided into a HIBI+BDNF group (group A),a HIBI group (group B) and a sham-operation group (group C). Models of HIBI were established by use of permanent ligation of the left common carotid artery followed by 2.5 hours ofinhalation of 8%O2+92%N2, then 0.5 μg BDNF was injected into the parietal cerebral ventricle in group A immediately. Contents of dynorphin A1-13-like, β-endorphin-like and leu-enkaphalin-like immunoreactivities (ir-DynA1-13, ir-β-EP and ir-LEK) in cortex and hippocampus were measured at 0, 60, 120 min after administration of BDNF. ResultsThe concentrations of ir-DynA1-13 and ir-β-EP in the cortex and hippocampus in group B were increased significantly than those in group C at most time points(P<0.05 or P<0.01). The contents of ir-LEK in left cortex and hippocampus in group B were significantly higher at 0 min after injury(P<0.05). After administration of BDNF, there were significant difference between the contents of ir-DynA1-13 in groups A and C at most time points except for those in hippocampus at 60 min and/or 120min(P<0.05 or P<0.01).The content of ir-β-Ep in the cortex in group A was significantly lower than that in group B at 60 min(P<0.05), while there was no significant difference between both groups at other time points. There was no significant difference between groups A and C with regard to the contents of ir-LEK in hippocampus. Conclusion Among 3 kinds of EOPs, ir-DynA1-13 and ir-βEp might be more closely related to HIBI. HIBI could be alleviated by exogenous BDNF with effects on Dynorphin and Endorphin systems.
