      目的研究脑缺血后不同时间开始亚低温对脑缺血损伤的影响。 方法脑缺血动物模型采用改良的Pulsinelli四动脉阻断法,48只大鼠随机分为假手术组(8只)、常温脑缺血组(8只)和脑缺血亚低温组(32只),后者再分为脑缺血后即时,30,60和90 min开始亚低温组,每组均为8只。观察不同时间开始亚低温对脑缺血组织抗氧化酶、MDA、ATP、乳酸和脑含水量的影响。 结果常温脑缺血组织中超氧化物歧化酶(SOD),谷胱甘肽过氧化物酶(GSH-Px),还原型谷胱甘肽(GSH)及三磷酸腺苷(ATP)的含量明显低于假手术组(P<0.01),丙二醛(MDA)、乳酸、含水量以及Ca2+的含量明显高于假手术组(P<0.01)。脑缺血后60 min内开始亚低温,可明显减少脑缺血组织中SOD,GSH-Px,GSH及ATP含量,而MDA、乳酸、含水量增加(P<0.05或P<0.01);脑缺血后90 min开始亚低温,组织中上述指标和常温脑缺血组差异无显著性意义(P>0.05)。 结论脑缺血后即时亚低温可明显延缓常温脑缺血组织中抗氧化酶及ATP的消耗,减少脂质过氧化产物和乳酸的堆积并减轻脑水肿。亚低温这种作用在脑缺血后30 min内实施效果好。

      Objective To study the effect of mild brain hypothermia beginning at different time after cerebral ischemia on cerebral ischemia injury. MethodsSprague-Dawley (SD) rats were treated with modified Pulsinelli 4-artery occlusion model, and randomized into a sham operation group, a normal temperature group and a mild hypothermia group. The later group were observed at 4 time points(mild hypothermia started at 0min, 30min, 60min and 90min after cerebral ischemia) with regard to the effects of mild hypothermia on SOD, MDA, ATP, lactate and water content. ResultsIn normothermia group, the amount of SOD,GSH-Px, GSH and ATP were lower and MDA, lactate, water content and Ca2+ was higher than those in the sham-operated group(P<0.05).Mild hypothermia beginning within 60min delayed the consumption of SOD, GSH-Px, GSH, ATP and decreased the accumulation of MDA, lactate, water content as compared with the normothermia group(P<0.05 or P<0.01). Mild hypothermia beginning at 90min resulted in no significant difference as compared with the normothermia group. ConclusionThe results showed that mild hypothermia alleviated ischemia injury when applied within 60min after cerebral ischemia.
