目的应用原子力显微镜(AFM)对电磁脉冲(EMP)辐射前、后培养的垂体细胞膜进行观察,以探讨EMP对其细胞膜影响的直接证据。 方法对Wistar大鼠垂体细胞在6孔板中进行原代培养,在培养第4天时,用高场强EMP模拟源(场强为6×104 V/m,脉冲上升时间为20 ns,脉宽为30 ms),以2.5次/min,照射2 min。并于照射后即刻固定细胞,应用日本岛津(SHIMADZU)公司的SPM-9500J3型原子力显微镜对细胞表面进行接触式连续扫描。 结果EMP辐射后即刻就可引垂体细胞膜表面大小不一、类圆形和不规则形的穿孔,穿孔最大口径和深度可达291 nm×372 nm×11.72 nm。 结论EMP辐射后可直接导致垂体细胞膜的穿孔,提示垂体细胞膜可能是EMP生物效应的靶部位之一。 |

Objective Using the atomic force microscope (AFM) to observe the membrane surface changes of pituitary cell of rat irradiated by electromagnetic pulse (EMP), and to explore the mechanism of injury of pituitary cell membrane caused by EMP. MethodsWistar rat's pituitary cell was collected and cultured in 6-hole plate. About 4 days later, the cells were exposed to EMP (with electric field intensity 60 kV/m, 20-nsec rise time and 30ms pulse wide) for 5 times in 2 min. The exposed cells were fixed immediately following irradiation and scanned by AFM (SPM-9500J3, SHIMADZU). ResultsThe results showed that there were different sizes and shapes of membrane perforate (the width is 291nm~372nm,the depth is 11.72nm) on the cell membrane. ConclusionEMP could cause the membrane perforate of pituitary cell. The membrane probably is the effect target of electromagnetic pulse. |
