目的研究局部热化疗对鼠胶质细胞移植瘤细胞增殖及PCNA、IGF-I表达的作用,探讨其作为胶质瘤辅助治疗的可行性。 方法将传代培养的C6细胞接种于大鼠背部皮下,肿瘤生长至直径为1.5~2.0 cm时,分组进行局部热疗、化疗和热化疗。继续观察皮下肿瘤生长情况,测量瘤体体积和重量;用免疫组化S-P法检测PCNA、IGF-I蛋白表达;用HE染色法、电镜观察肿瘤增殖状况。 结果热疗、化疗、热化疗与对照组比较,瘤体缩小,瘤重减轻(分别为2.95,1.95,1.86和1.09 g,P值均<0.01);PCNA蛋白和IGF-I蛋白表达降低(P<0.01),且热化疗较单纯热疗和化疗降低更明显(P<0.01)。出现核染色质凝聚,凋亡小体,细胞增殖明显受抑制。 结论①热疗、化疗、热化疗能抑制肿瘤增殖,且热化疗有协同作用。②热疗、化疗、热化疗能减低PCNA、IGF-I蛋白表达。③热化疗抑制胶质瘤的PCNA合成与表达,抑制IGF-I的合成与分泌,是其治疗肿瘤的机制之一。 |

Objective To study the effects of localized thermochemotherapy on cellular proliferation and the expression of PCNA and IGF-I protein of C6 gliomas in rat, and explore the possibility of developing new therapeutic method of gliomas. MethodsC6 glioma cells were injectcd subcutaneously to rats. The rats were randomly assigned to 4 groups,and treatment were initiated on day 22 after tumor inoculation. Prior to treatment, the subcutaneous gliomas were examined to verify tumor size, which were ranged 1.5 to 2.0 cm. Then localized thermochemotherapy, hyperthermia and chemotherapy were given, respectively. The size and weight of subcutaneous tumors were measured. PCNA and IGF-I protein expression were detected by the technique of S-P immunohistochemistry. Glioma cells proliferation was detected by HE staining method and electron-microscopic observation. ResultsCompared with the control group, the tumor size of rats in the hyperthermia and chemotherapy as well as thermochemotherapy groups was decreased,the tumor weight was also decreased significantly (2.95g vs 1.95g vs 1.86g vs 1.09g,P<0.01), while the expression of PCNA and IGF-I protein were decreased. Electron-microscopic examination showed that proliferation was inhibited by interstitial localized thermochemotherapy in glioma rat. ConclusionHyperthermia,chemotherapy and thermochemotherapy could inhibit the growth of subcutaneously inoculated glioma cells. Adriamycin( ADM) has marked synergic effects with hyperthermia in C6 glioma of rat; Hyperthermia and chemotherapy as well as thermochemotherapy can decrease the expression of PCNA and IGF-I protein; Proliferation of glioma cells can be inhibited by localized thermochemotherapy and may be one of the mechanism to explain the decreased expression of PCNA and IGF-I protein. |
