目的探讨高压氧(HBO)对持续性局灶性脑缺血细胞凋亡的影响及其作用机制。 方法选择4~5个月龄SD大鼠65只,随机分为缺血组、HBO组和假手术组。观察脑梗死灶体积、凋亡细胞数量及凋亡调控基因蛋白表达的变化。 结果HBO组与缺血组相比,6 h~10 d各相应时间点梗死灶体积均明显缩小,差异均有极显著性意义(P均<0.01);HBO组缺血48 h后,TUNEL阳性凋亡细胞数较缺血组明显减少(P<0.01);HBO组与缺血组相比,在缺血6 h~10 d各时间点Bcl-2阳性细胞增加,差异有显著性意义;Bax阳性细胞在缺血6 h~10 d各时间点均减少,差异有显著性意义。 结论脑缺血早期暴露于高压氧可以减少局灶性脑缺血梗死灶体积;高压氧能抑制脑缺血细胞凋亡,其作用与上调Bcl-2蛋白、下调Bax蛋白表达有关。 |

Objective To study the effect of hyperbaric oxygenation (HBO) on apoptosis in focal cerebral ischemia and the mechanisms underlying it. MethodsSixty five 4-5-month-old Spraque-Dawley(SD)rats were randomly divided into three groups: the focal cerebral ischemia group,FCI group,HBO group and the control group.The focal brain tissue infarct sizes(BTIS),the number of apoptotic cells and the expression of bcl-2 and Bax protein were examined at different time points. ResultsThe BTIS were significantly reduced from 6 hours to 10 days, the number of apoptotic cells were significantly decreased in 48 hours after HBO in the HBO group (P<0.01). The bcl-2 protein levels were increased significantly and Bax protein levels decreased significantly in HBO group (all P<0.01). ConclusionHBO can reduce focal brain tissue infarct size and inhibit apoptosis when administered in early stage of focal cerebral ischemia. This may be related to the up-regulation of bcl-2 protein levels and down-regulation of Bax protein levels. |
