目的比较褪黑激素和甲泼尼龙对实验性脊髓损伤的保护作用。方法24只SD大鼠随机分4组,一组椎板切除不损伤脊髓,另三组用Allen法中度量(50 g•cm)损伤脊髓,分别给予褪黑激素每公斤体重100 mg 腹腔注射;甲泼尼龙每公斤体重30 mg腹腔注射和含5%乙醇的生理盐水腹腔注射,4 h后取伤段脊髓,测量组织的游离铁,丙二醛(MDA)和谷胱甘酞过氧化物酶(GSH px)活性。结果损伤后4 h伤段脊髓组织的游离铁有明显的升高,导致MDA大量生成,组织GSH px活性升高;褪黑激素和甲泼尼龙能降低脊髓损伤后游离铁的释放(P<0.05),减少MDA的生成(P<0.05),抑制组织GSH px活性升高(P<0.05)。结论褪黑激素和甲泼尼龙能同样减轻脊髓损伤后游离铁浓度升高触发的氧化作用和脂质过氧化反应(P<0.05)。 |

ObjectiveTo observe the protective effects of melatonin and methylprednisolone on acute experimental spinal cord injury in rats. MethodsTwenty four Sprague Dauleys(SD) rats were randomly divided into four groups, laminectomy were performed in rats in group one and nontraumatized spinal cord samples were obtained. The spinal cord of rats in other groups were injured by Allen's weight drop method, then a single dose of melatonin(100 mg/kg), a single dose of methylprednisolone(30 mg/kg), and saline containing 5% ethanol were administered intraperitoneally. The levels of free iron, MDA and GSH px of the involved spinal cord segments were measured 4 hours after the injury. ResultsIncreased levels of free iron with resultant massive production of MDA, and elevated activities of tissue GSH px were observed 4 hours after the spinal cord injuries in rats administered with melatonin or methylprednisolone injection. The production of free iron was limitted and both of the activities of MDA and GSH px would be significantly suppressed (P<0.05). ConclusionThis experimental results demonstrated that both melatonin and methylprednisolone can exert protective action on spinal cord injuries |
