目的观察关节腔注射羧甲基壳聚糖(CMCTS)对骨关节炎(OA)模型中关节软骨退变及软骨中基质金属蛋白酶-1,-3(MMP-1,-3)mRNA表达的影响。 方法16只大耳白兔行单侧前交叉韧带切断术,术后5周将动物随机分为2组,每组8只,实验组关节腔注射2%CMCTS 0.3 ml,每2周1次,共注射3次,对照组不注射任何药物。术后11周处死动物,比较各组股骨内髁关节软骨的大体变化,采用RT-PCR方法检测软骨中MMP-1和MMP-3的mRNA表达。 结果实验组软骨退变程度较对照组明显减轻,实验组软骨MMP-1和MMP-3的mRNA表达明显低于对照组(P<0.01)。 结论关节腔注射CMCTS能够明显抑制OA软骨中MMP-1和MMP-3 mRNA的表达,明显减轻软骨退变的程度,对软骨有一定保护作用。 |

Objective To observe the influence of intra-articular injection of carboxy-methylated chitosan (CMCTS) on cartilage degradation and mRNA expression of matrix metalloproteinase-1 (MMP-1) and matrix metalloproteinase-3 (MMP-3) of cartilage in osteoarthritis (OA). MethodsSixteen white rabbits underwent experimental unilateral anterior cruciate ligament transection (ACLT) were divided into an experiment group and a control group. Five weeks after the operation, the rabbits in the experiment group were administered with intra-articular injection of 0.3ml of 2% CMCTS, once two weeks, while those in the control group were not injected after ACLT. All the animals were sacrificed 11 weeks after surgery, and the changes of the cartilage on the medial femoral condyles were compared between the 2 groups. The mRNA expressions of MMP-1 and MMP-3 were analyzed using reverse transcription-polyme-rase chain reaction (RT-PCR). ResultsCartilage degradation in the control group was significantly more severe than that in the experiment group. The cartilage mRNA expression of MMP-1 and MMP-3 was suppressed in the CMCTS injection group. ConclusionCMCTS could significantly decrease the mRNA expression of MMP-1 and MMP-3 and reduce the severity of cartilage degradation. CMCTS has a protective effect on articular cartilage. |
